Zepbound® (tirzepatide) is a once-weekly injectable prescription medication. Tirzepatide acts on receptors for two naturally occurring hormones involved in metabolic regulation: glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1).
FDA-approved uses
Zepbound is FDA-approved, together with a reduced-calorie diet and increased physical activity, to reduce excess body weight and maintain weight reduction long term in adults with obesity or adults with overweight who have at least one weight-related condition.
It is also FDA-approved to treat moderate-to-severe obstructive sleep apnea in adults with obesity.
How it works

Wegovy® injection (semaglutide) is a once-weekly injectable prescription medication. Semaglutide is a glucagon-like peptide-1 (GLP-1) receptor agonist.
FDA-approved uses
Wegovy is FDA-approved, together with a reduced-calorie diet and increased physical activity, to reduce excess body weight and maintain weight reduction long term in adults with obesity or adults with overweight who have at least one weight-related condition, and in certain pediatric patients with obesity.
Wegovy is also FDA-approved to reduce the risk of major adverse cardiovascular events in adults with established cardiovascular disease and either obesity or overweight.
Wegovy injection is additionally FDA-approved for the treatment of noncirrhotic metabolic dysfunction-associated steatohepatitis (MASH) with moderate-to-advanced liver fibrosis in adults. This indication was granted under accelerated approval.
How it works

Zepbound® (tirzepatide) has a boxed warning about the risk of thyroid C-cell tumors. In studies in rats, tirzepatide caused thyroid C-cell tumors. It is not known whether Zepbound causes thyroid C-cell tumors, including medullary thyroid carcinoma (MTC), in humans.
Do not use Zepbound if you or a family member have had MTC, or if you have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). Do not use Zepbound if you have had a serious allergic reaction to tirzepatide or any of its ingredients.
Zepbound may cause serious side effects, including severe gastrointestinal reactions, acute kidney injury due to volume depletion, gallbladder problems, acute pancreatitis, serious allergic reactions, low blood sugar, particularly when used with insulin or certain diabetes medicines, and diabetic retinopathy complications in patients with type 2 diabetes. Zepbound delays gastric emptying, and rare cases of pulmonary aspiration have been reported in patients receiving GLP-1-based medicines during procedures requiring general anesthesia or deep sedation. Tell your healthcare team that you take Zepbound before planned surgery or procedures.
Zepbound is not recommended in patients with severe gastroparesis. Tell your healthcare provider if you are pregnant, plan to become pregnant, or take other prescription or over-the-counter medicines.
The most common adverse reactions reported with Zepbound include nausea, diarrhea, vomiting, constipation, abdominal pain, indigestion, injection-site reactions, fatigue, hypersensitivity reactions, belching, hair loss and gastroesophageal reflux disease.

Patients using oral hormonal contraceptives should discuss contraception with their provider because the prescribing information recommends switching to a non-oral contraceptive method or adding a barrier method for 4 weeks after starting Mounjaro and for 4 weeks after each dose increase.

Mounjaro and Ozempic are FDA-approved for type 2 diabetes and have additional FDA-approved indications in specified populations, but they are not FDA-approved for chronic weight management.
